Linda B. Buck ist eine Neurophysiologin, die am 29. Januar 1947 in Seattle, Washington, geboren wurde. Gemeinsam mit Richard Axel erhielt sie 2004 den Nobelpreis für Physiologie oder Medizin für ihre wegweisende Forschung zum Riechsystem.
Bucks wissenschaftliche Karriere führte sie von der Harvard Medical School zur University of Washington in Seattle, wo sie seit 2003 als Professorin für Physiologie tätig ist. Zusätzlich ist sie in der Basic Sciences Division am Fred Hutchinson Cancer Research Center engagiert und forscht am Howard Hughes Medical Institute als Professorin für Physiologie und Biophysik.

Bucks Forschung konzentriert sich auf die Übertragung von Reizen an das Gehirn, insbesondere auf olfaktorische Reize. Sie untersucht die Entstehung und Entwicklung von Geruchsrezeptoren sowie die Verarbeitung von Gerüchen im Gehirn und deren Auswirkungen auf Reaktionen, Gedanken und Verhalten. Ihre Arbeitsgruppe identifizierte eine Genfamilie von etwa 1000 Genen, die mit der Geruchswahrnehmung in Verbindung stehen und für verschiedene Geruchsrezeptoren codieren. Diese Rezeptoren befinden sich in der Riechschleimhaut und sind direkt mit dem Bulbus olfactorius verbunden, dem Gehirnbereich für die Geruchswahrnehmung.
Axel und Buck konnten unabhängig voneinander zeigen, dass jedes Neuron nur einen Rezeptortyp anspricht und dass die Rezeptoren in der Riechschleimhaut zufällig verteilt sind, aber im Bulbus olfactorius in derselben Region wahrgenommen werden. Auf diese Weise entsteht im Gehirn eine komplexe Geruchsempfindung. Neben der Erforschung des Geruchssinns widmete sich Buck auch der Erforschung des Alterns und der genetischen Beeinflussung der Lebensspanne am Beispiel des Fadenwurms Caenorhabditis elegans.
In den Jahren 2008 und 2010 zog Buck drei wissenschaftliche Arbeiten zurück, nachdem sie und ihre Mitarbeiter die Ergebnisse eines ehemaligen Mitglieds ihrer Arbeitsgruppe nicht reproduzieren konnten, wobei die Nobelpreisverleihung auf anderen Arbeiten basierte. Im Laufe ihrer Karriere wurde Linda Buck mit zahlreichen Preisen und Ehrungen ausgezeichnet, darunter das Sloan Research Fellowship (1992), der Rosenstiel Award (1996) und die Mitgliedschaft in der National Academy of Sciences (2003).

Linda Buck’s research has transformed the field of odorant perception through the cloning of mammalian olfactory receptors.
